                Microorganisms which are ubiquitous and have the tendency to adapt quickly to their immediate environment are responsible for considerable mortality and morbidity worldwide especially in developing countries due to poor sanitation, unhygienic and overcrowded living conditions (Theuretzbacher  2011). Various studies have established that herbal medicines can be developed as safe, effective and less costly alternatives to the current medicines to the treat certain infections (Vermani and Garg 2002).      

                This study was conducted to determine the antibacterial activity of secondary metabolites of Bixa orellana Linn. (1992) leaf decoction against Bacillus subtilis and  Escherichia coli using Thin Layer Bioautographic Overlay Assay. The solvent system was used methanol: chloroform (85:15) together with the pre-coated TLC plate. Amoxicillin, 500 mg capsule, was dissolved in sufficient distilled water make 25 mL positive control solution with a concentration of 20 mg/mL. The negative control was distilled water. The bacterial cell density was 1.5 x 107 and the medium was Mueller-Hinton agar and Nutrient broth.  

                Three spots were observed on the chromatogram. Spot A with Rf value 0.017 Spot B with Rf value 0.629 and Spot C with Rf value 0.700. Clear, white zones were seen in the Overlay Assay with Spot B and Spot C and none with Spot A. Results were positive with Antimony (III) Chloride, Potassium Ferricyannide-Ferric Chloride and Vanillin-Sulfuric acid indicating the possible presence of flavonoids, tannins and steroids.      

                If the clear, white zones considered zone of inhibition against Eschericha coli then it is concluded that the leaf decoction of Annatto is a weak antibacterial agent due to its low antibacterial activity compared to the positive control.
