In vitro hypoglycemic activity of Blumea balsamifera Linn. 1833 (Gabon) leaf extract by inhibition of a-amylase and a-glucosidase enzymes.

The hypoglycemic activity of the Blumea balsamifera Linn. 1833 (Gabon) leaf extract was determined by α-amylase and α-glucosidase inhibitory activity assay. Three solvents were used for the extraction; distilled water for decoction, 95% ethanol, and dichloromethane for maceration. The absorbance of the by-products in α-amylase and α-glucosidase assay are 3-amino-5-nitrosalicylic  acid, a red-orange solution was measured at 540nm, and p-nitrophenol, a yellow solution, was  measured at 540nm, p-nitrophenol, a yellow solution, was measured at 405nm respectively. Decrease in concentration resulted to decrease in absorbance, making the percent (%) inhibition and percent (%) hypoglycemic activity increased. For α-amylase assay with 0.075mg/mL concentration, distilled water-free test solution has the  highest percent hypoglycemic activity at 97.48%, followed by ethanol-free test solution at 72.49% then dichloromethane-free test solution  at 54.82%. For α-glucosidase assay, results showed that with a concentration of 0.075mg/mL, distilled water-free test solution has the highest percent hypoglycemic activity at 95.66%, followed by ethanol-free test solution at 71.49% then dichloromethane-free test solution at 51.48%. The hypoglycemic activity of  the test extract of B. balsamifera could possibly be due to the presence of saponins, tannins, phenolic compounds and terpenes. The researchers recommend that further investigation be done on the isolation, purification and characterization of the chemical constituents specifically total phenolic content and saponins which are responsible for the hypoglycemic activity found in the leaves of Blumea balsamifera. It is also recommended that in α-amylase assay, the temperature used in boiling the test tubes must be constant in all extracts to compare the activity per extract. It is also recommended for a new method of assay and enzymes other than α-amylase and α-glucosidase such as lipase, sucrose and glucoamylase to further find out if the test extracts of B. balsamifera will exhibit hypoglycemic activity.
