The anti-arthritic potential of Barbados Nut Jatropha curcas Linn. (1753) leaf extract in bovine type II collagen-induced arthritis (CIA) in male albino mice Mus musculus.

Rheumatoid arthritis (RA) is a common autoimmune systemic inflammatory disease affecting approximately 1% of the worldwide population (Gibofsky, 2012). In 2008, the Department of Health has estimated about 11 Million Filipinos with Osteoarthritis (based on a prevalence rate of 16%) and about 50% of 65 years and older with symptoms of arthritis . Many plants have been studied for their anti-arthritic and anti-inflammatory effects; some are documented but most are folkloric accounts. J. curcas have been widely used in  oriental clinics to treat rheumatism and arthritis as an effective alternative by traditional users (Salimon and Abdullah, 2008; Kaushik et. al.,2007). Thus study aimed to determine the anti-arthritic potential of the extract of barbados  nut leaf extract (Jatropha curcas L. 1753) on bovine type II collagen-induced arthritis (CIA) in male albino mice (Mus musculus). The plant extract was tested for its anti-arthritic potential using three different doses: these were 250mg/Kbw (A), 500mg/Kbw (B), and 750mg/Kbw (C). The positive control used was the acetic acid derivative NSAID, Indomethacin and the negative control was distilled water. The results of the experiment showed that the plant extract exhibited  a anti-arthritic effect on bovine type II collagen induced arthritis (CIA) in male albino mice. Among the different doses, it was the 750mg/Kbw dose that exhibited the highest average percent reduction with 128.33% followed by 500mg/Kbw dose with 120.37% then the 250mg/Kbw dose with 96.76%. However, the anti-arthritic activity of the test doses was not comparable to the positive control which gave 288.89% paw swelling reduction. A two-way analysis of variance (p value.0.01) was used to compare the percent activity and percent reduction. In the post hoc analysis (p value less than 0.01), the comparison of the different doses shows that the percent anti-arthritic activity are negligible among the three test doses but comparable to that of the positive control. Using linear regression analysis, the median effective dose (ED50) is 963.86 mg. moreover, results show that the dose is directly proportional to the percent anti-arthritic activity, this would mean that the activity of the leaf extract of Jatropha circas Linn is dose-dependent.
