Anticoagulant activity of the leaf extract of Pigweed, Amaranthus spinosus Linn. (1753) using modified lee-white method.

The aqueous extract of dried leaves of Amaranthus spinosus (Fam: Amaranthaceae) was tested for their anticoagulant activity on human blood (type O+) by Modified Lee-White method. The different test solutions were placed in different test tubes and freshly collected blood was added. They were then placed in a water bath at 370C. The time was recorded when the test tubes were already placed in the water bath until a blood clot has formed. The 25% saturated solution of the leaf extract of A. spinosus significantly increased clotting time (p-value is 0.00000232) compared to the untreated blood. The 100% (6.4mg/mL) saturated solution exhibited 6.74% anticoagulant activity and the 50% (3.2mg/mL) saturated solution exhibited 23.63% anticoagulant activity, while the 25% (1.6mg/mL) saturated solution exhibited 54.24% anticoagulant activity. From these observations, it is confirmed that the aqueous extract of Amaranthus spinosus. The treatment groups were left to stand for 12 hours to be examined for clot retraction. All the retreatment groups had a complete clot retraction except for the positive control which is heparin. This is expected for heparin because it is already known to have a side effect that delays the rate of clot retraction, it is called heparin-induced thrombocytopenia. The reason why the 50% and 100% saturated solutions had a lower activity is due to the presence of calcium in the A. spinosus leaf extract. The calcium present in the 25% saturated solution was not enough to catalyze the coagulation of the blood, while at the higher concentrations, the amount of calcium present decreased the time needed to form a blood clot. Based on the results of the research, the researchers have concluded that the A. spinosus leaf extract has anticoagulant activities.
